Sciences"}]},"item_12_description_4":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"細胞DNA量をカラー画像解析装置を用いて顕微測光する方法を考案した.本法は,基本的には従来の顕微分光測光法と同様の原理(Lambert-Beerの法則)に基づくものであるが,顕微鏡・カメラを介して入力された画像を用いて測定を行うため,顕微分光測光法とは大きく異なる点がある.また,核DNAの染色物質に特異的な干渉フィルター(band-pass filter)を用いる代わりにカラーカメラの映像を構成する三原色成分の画像を利用することも本法のユニークな点である.本研究では,まず,今回考案した画像解析法の理論的検討を行った.その要点は,分光測光法の最大の問題である分布誤差と非特異的光喪失への対処方法にある.すなわち,1)顕微分光測光法では,分布誤差を減ずるために測定領域を小さな測光スポットで走査する方法が用いられている.画像解析法では,顕微鏡を介して撮影された画像を用いて測定を行うが、画像は画素(pixel)と呼ばれる小さな区画の集合であり,測定対象は入力された時点分割(ディジタル化)されるので,走査の必要がない.顕微分光測光法が「点」での測定に対して,画像解析法では「面」での測定となるため分布誤差の減少,測定精度の向上が期待でき,また,測定時間の短縮が図れる.2)非特異的光喪失に関しては,分光測光法では通常,干渉フィルターを用いた二波長計測することで対処している.本画像解析法においては撮像装置がカラー対応であることに着目し,より簡便に非特異的光喪失を除去する方法として,カラー画像を構成する二成分を干渉フィルターの代わりに利用する方法を考察し,それが理論的に可能であることを示した.(第2章)次に,この細胞核DNAのカラー画像解析法の理論に基づいて実際の測定を行うために必要な装置の構成及び,測定解析のアルゴリズム・手順を具体的に検討した.その際,重要な点はカラーカメラとして自己走査型荷電結合素子(CCD;Charge-Coupled Device)カメラを用い,測定解析アルゴリズムを一般に普及している汎用画像処理装置で稼動できるものとした点である.引き続いて,実際に非特異的喪失除去するための係数γを決定し,各種細胞核DNA量の測定を行い,本画像解析法の定量性・精度を検討した.具体的には,1)本測定法の系統誤差を評価するために,同じDNA量を持ちながら,核の形態・面積が異なるリンパ球と好中球のDNA量を測定し,両者の値を比較した.2)本法の定量性を検討するために,リンパ球の核DNA量とその半分のDNA量を持つDNA量を測定した.3)光波長フィルター(カラーカメラの色成分)を利用したことの妥当性を調べるために,干渉フィルター(band-pass
